UAS regulatory sequences drive expression of Hsap\SNCA. This construct was generated using the P{Express-UAS} vector.
Expression of Hsap\SNCAScer\UAS.cJa under the control of Scer\GAL4ninaE.PT results in age-progressive degeneration of ommatidia as well as vacuolar lesions in the adult retina.
Expression of Hsap\SNCAScer\UAS.cJa driven by Scer\GAL4ninaE.PT (crosses carried out at 18[o]C and shifted to 25[o]C following eclosion) does not significantly affect external eye morphology in adult flies. At 30 days old (but not 1 or 10), electroretinogram (ERG) recordings of light induced potentials from Scer\GAL4ninaE.PT>Hsap\SNCAScer\UAS.cJa flies show a significant decrease in light-evoked in depolarization amplitude compared to controls; mutant flies show significant increases in ON transient amplitude at day 10 and 30 and significant decreases in OFF transient amplitude at day 10 and 30 (no differences at day 1).
The number and morphology of photoreceptors (retinae show intact ommatidia with 7 visible rhabdomeres) in Scer\GAL4ninaE.PT>Hsap\SNCAScer\UAS.cJa flies is grossly preserved at day 1, similar to controls; with aging (day 10 and 30), progressive vacuolar changes affect the retina and there is a significant decrease in rhabdomere numbers at day 10 (at day 30 the severity of tissue loss and degeneration is such that ommatidia cannot be reliably identified). Photoreceptor ultrastructure (examined with TEM) in 10 day old Scer\GAL4ninaE.PT>Hsap\SNCAScer\UAS.cJa flies revealed significant subtle defects and irregularities in rhabdomeres (splitting or fraying at edges, vacuolar changes). Mutants at day 10 have similar numbers of mitochondria per ommatidium and percentage of photoreceptors with electron dense vacuoles (telolysosomes) as controls, but have a significant increase in the number of autophagic vacuoles (increased numbers of multivesicular or multilamellar bodies). Scer\GAL4ninaE.PT>Hsap\SNCAScer\UAS.cJa flies show significant decrease in photoreceptor synaptic terminal area and synaptic vesicle density at 10 days old compared to controls.

The age-progressive retinal degeneration characteristic for flies expressing Hsap\SNCAScer\UAS.cJa under the control of Scer\GAL4ninaE.PT is significantly enhanced by co-expression of any of the following: CG32191GD6163, CG32191KK103889, CG12025KK107653, CG12025GD2253, Ptp10DKK101775, Ptp10DGD115, Ptp10DGD2611, Vps13HMS01715 and Vps13HMS02460 or by combination with Ptp10D1 and Ptp4E1 in double heterozygous state, combination with single copy of each allele alone does not exacerbate the retinal phenotype.
