Expressing SLC22AKK112539 under the control of Scer\GAL4fkh.PH does not significantly affect the clearing of salivary gland tissue in pupae, as compared to controls.

Expression of SLC22AKK112539 (with UAS-Dicer2 to enhance RNAi efficiency) either pan-neuronally (Scer\GAL4nSyb.PU), in all mushroom body (MB) neurons (using one of Scer\GAL4238Y, Scer\GAL4ey-OK107, Scer\GAL4GMR13F02 or Scer\GAL4c747) or in dopamine neurons (using Scer\GAL4ple.PU) results in significant enhancement of 3hr memory in an olfactory classical conditioning assay in adult flies. A significant enhancement of memory is also seen when expression is driven by either Scer\GAL4c305a (α'/β' MB neurons) or Scer\GAL4GMR28H05 (α/β MB neurons), but not when driven by Scer\GAL41471 (γ MB neurons).

Expression of SLC22AKK112539 (with UAS-Dicer2 to enhance RNAi efficiency) in the glia (using one of Scer\GAL4repo.PU or Scer\GAL4nrv2.PU) has no significant effect on 3hr memory in an olfactory classical conditioning assay in adult flies.

Expression of SLC22AKK112539 (with UAS-Dicer2 to enhance RNAi efficiency) has no significant effect on 3 hour memory in an olfactory classical conditioning assay in adult flies when expression is driven by any one of Scer\GAL4VT064246, Scer\GAL4GH146, Scer\GAL4Mz604 or Scer\GAL4NP2492.

Odor (Benzaldehyde and Octanol) and shock (electrical) avoidance was normal (tested with Scer\GAL4238Y, Scer\GAL4GMR13F02, Scer\GAL4c747, Scer\GAL4ey-OK107, Scer\GAL4c305a, Scer\GAL4GMR28H05, Scer\GAL4nSyb.PU, Scer\GAL4ple.PU).

A conditional knock-down of SLC22A after eclosion in all MB neurons using the Scer\GAL4238Y driver (with UAS-Dicer2 to enhance RNAi efficiency and GAL80ts to limit the stage of expression) results in enhanced 3hr memory in an olfactory classical conditioning assay in adult flies whereas knock-down during development (before eclosion) has no effect on memory test performance in adults.

Expression of SLC22AKK112539 RNAi in all MB neurons (using one of the following drivers Scer\GAL4238Y, Scer\GAL4GMR13F02 with UAS-Dicer2 to enhance RNAi efficiency) results in significant memory acquisition impairment in an olfactory classical conditioning assay in adult flies. Memory acquisition impairment is also seen when expression is driven in the α' and β' MB neurons using the Scer\GAL4c305a driver (with UAS-Dicer2 to enhance RNAi efficiency).

Expression of SLC22AKK112539 RNAi in all MB neurons using the Scer\GAL4238Y driver (with UAS-Dicer2 to enhance RNAi efficiency) results in significant enhancement of memory retention in in an olfactory classical conditioning assay in adult flies.

Adult SLC22A knock-down flies (expressing SLC22AKK112539 RNAi in all MB neurons using the Scer\GAL4238Y driver with UAS-Dicer2 to enhance RNAi efficiency) showed enhanced memory retention in an olfactory classical conditioning assay after cold-shock compared to controls.

Expression of SLC22AKK112539 RNAi in all MB neurons using the Scer\GAL4238Y driver (with UAS-Dicer2 to enhance RNAi efficiency) does not change the rate of memory decay in an olfactory classical conditioning assay in adult flies.

Adult SLC22A knock-down flies (expressing SLC22AKK112539 RNAi in all MB neurons using the Scer\GAL4238Y driver with UAS-Dicer2 to enhance RNAi efficiency) showed greater interference in a proactive interference memory test in an olfactory classical conditioning assay compared to controls.

Adult SLC22A knock-down flies (expressing SLC22AKK112539 RNAi in all MB neurons using the Scer\GAL4238Y driver with UAS-Dicer2 to enhance RNAi efficiency) showed lesser interference in a retroactive interference memory test in an olfactory classical conditioning assay compared to controls.

Supplementing food with dopamine or histidine impairs 6hr memory in a in an olfactory classical conditioning assay in control adult flies and memory-enhanced flies expressing SLC22AKK112539 RNAi under the Scer\GAL4238Y driver (with UAS-Dicer2 to enhance RNAi efficiency) to the same degree.

Supplementing food with choline improves 6hr memory in an olfactory classical conditioning assay in control adult flies but has no further effect on the memory-enhanced flies expressing SLC22AKK112539 RNAi under the Scer\GAL4238Y driver (with UAS-Dicer2 to enhance RNAi efficiency).
