Ectopic expression of MitfKK113614 RNAi under the control of Scer\GAL4ey-OK107 results in a significant accumulation of polyubiquitinated protein aggregates and the expression under Scer\GAL4elav.PU leads to significantly increased size of mitochondria in the brain of adult flies upon starvation.

The fat body of third instar larvae expressing MitfKK113614 RNAi under the control of Scer\GAL4[Lsp2.PH]) shows accumulation of the ref(2)P protein regardless of their fed/starved status.

Ectopic expression of MitfKK113614 RNAi under the control of Scer\GAL4Lsp2.PH results in a significantly decreased number of acidic vesicles in the fat body of starved third instar larvae, while the number of lysosomes is significantly increased in fat bodies of fed but not starved larvae compared to controls. MitfKK113614 RNAi expression under also prevents normal accumulation of lipid droplets in fed larvae: both fed and starved larvae display the same (low) amount of lipid droplets as starved wild-type controls.

Ectopic expression of MitfKK113614 RNAi under the control of Scer\GAL4elav.PU results in mild accumulation of lysosome-like structures and autophagosomes in the brains of fed adult flies compared to controls and leads to increased size of lysosome-like structures and the appearance of lamellar bodies as well as significantly decreased number of autophagosomes in the brains of starved flies compared to starved controls.

Ectopic expression of MitfKK113614 RNAi under the control of Scer\GAL4Lsp2.PH results in significantly decreased level of lysosome-autophagosome fusion in starved third instar larvae compared to starved wild-type but the fusion level in fed larvae compared to fed controls is unchanged.

Expression of MitfKK113614 under the control of Scer\GAL4byn-Gal4 leads to early larval lethality.

Comments

The early larval lethality of animals expressing MitfKK113614 under the control of Scer\GAL4byn-Gal4 is rescued by co-expression of MitfSI1-RES.
