Atg16MI00187 homozygous adult guts show severe morphological changes, including significant increases in length and diameter accompanied by a significantly thicker epithelium, as compared to controls; this mutant epithelium shows a significant increase in the proportion of pre-enteroendocrine cells and a reciprocal significant decrease in the proportion of mature enteroendocrine cells, associated with a significant delay in pre-enteroendocrine cell maturation and a significant increase in the overall number of Prospero-positive precursor/mature enteroendocrine cells, but shows no changes in the thickness of the peritrophic membrane, in the proportion of mitotic or apoptotic cells or in cell adhesion, as compared to controls.

Atg16MI00187 homozygous adults show a severe decrease in lifespan and a significant increase in the gut bacteria burden upon oral infection with Pseudomonas aeruginosa or Enterococcus faecalis, as compared to control; in the absence of infection, these individuals show a significant increase in the gut burden of Enterobacteriaceae (but not in the overall burden of bacteria, Lactobacilli or Acetobacteriaceae), as compared to controls. These mutants do not show significant changes in food uptake.

Atg16MI00187/Atg16MI00187 mutants have similar spermiogenesis to wild type, with spermatid nuclei showing wild-type morphology.

Both the increased proportion of pre-enteroendocrine cells and the reciprocal decreased proportion of mature enteroendocrine cells observed in the adult gut of Atg16MI00187 are fully suppressed by the expression of robo2EP2582 or sliEY10695 under the control of Scer\GAL4esg.PU; their severe decrease in longevity upon oral infection with Pseudomonas aeruginosa or Enterococcus faecalis is also suppressed by the expression of sliEY10695 or robo2EP2582 under the control of Scer\GAL4esg.PU.

    Comments

Both the increased proportion of pre-enteroendocrine cells and the reciprocal decreased proportion of mature enteroendocrine cells observed in the adult gut of Atg16MI00187 are fully suppressed by the expression of Atg16Scer\UAS.c.T:Zzzz\FLAG, Atg16ΔAutD.Scer\UAS or Atg16ΔAutD+Linker.Scer\UAS, but not Atg16ΔWD40.Scer\UAS, under the control of Scer\GAL4esg.PU.

The severe decrease in longevity of Atg16MI00187 homozygotes upon oral infection with Pseudomonas aeruginosa or Enterococcus faecalis is suppressed by the expression of Atg16ΔAutD+Linker.Scer\UAS under the control of Scer\GAL4esg.PU.
