Expression of nompCL.Scer\UAS under the control of Scer\GAL4GH298 in antennal lobe interneurons renders these interneurons sensitive to mechanical stimulation (these neurons show no mechanosensitivity in wild-type controls). Inward currents are recorded at the onset and the end of the stimulus in the mutant interneurons. The latency of the major 'on' current is less than 5 ms and the amplitude correlates with the distance of stimulating probe displacements.
Expression of nompCL.Scer\UAS in interneurons under the control of Scer\GAL4kra-krasavietz causes these interneurons to become sensitive to mechanical stimulation.
Unlike in wild type, class IV dendritic arborization neurons expressing nompCL.Scer\UAS under the control of Scer\GAL4ppk.PH produce a burst of action potentials and an increase in internal calcium level in response to touch stimulation.
Scer\GAL419-12/nompCL.UAS rescues nompC1/nompC3
nompCL.UAS/Scer\GAL4221 rescues nompC1/nompC3
Expression of nompCL.Scer\UAS under the control of Scer\GAL4nompC.PC rescues the reduced locomotion of nompC1/nompC3 adults.
Expression of nompCL.Scer\UAS under the control of Scer\GAL419-12 rescues the touch sensitivity defects seen in nompC1/nompC3 mutant larvae. The defects in behavior, action potential frequency and calcium response are all rescued.
The lethality of nompC1/nompC3 larvae is rescued by expression of nompCL.Scer\UAS under the control of Scer\GAL4nompC.PC. The reduced crawling speed and increased stride duration seen in nompC1/nompC3 larvae is also rescued.
The reduced crawling speed and increased stride duration of nompC1/nompC3 larvae is rescued by expression of nompCL.Scer\UAS under the control of either Scer\GAL45-40, Scer\GAL421-7 or Scer\GAL42-21.
The walking defects seen in nompC1/nompC3 adults are rescued by expression of nompCL.Scer\UAS under the control of either Scer\GAL421-7 or Scer\GAL4nompC.PC.
