UASp regulatory sequences drive expression of a mutated form of gro, in which alanine residues replace the phospho-acceptor residues within the two rl consensus sites of gro, rendering it unphosphorylatable.

The expression of groAA.Scer\UAS.P\T under the control of Scer\GAL4109-30, in combination with gal80[ts] to restrict expression to adulthood, leads to basal stalk defects, including increased number of basal stalk cells and multilayering or elongation in the majority of cases, as compared to controls; clonal follicle stem cells expressing groAA.Scer\UAS.P\T under the control of Scer\GAL4tub.PU exhibit a significant decrease in cell competition capacity, as seen by their increase in clonal extinction as compared to wild-type control clones.

Maternal expression of groAA.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 results in 86% loss of the intermediate neuroblast column of the neuroectoderm in embryos.

Maternal expression of groAA.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 results in a low hatching rate. 30.5% of unhatched larvae show strong reduction or even loss of the filzkorper.
