    Detailed Description

At stage 17, 37% of homozygous embryos show inversion of the normal left-right asymmetry of the anterior midgut, 14% do not show any left-right asymmetry and 49% show normal left-right asymmetry. In addition the valvular structure of the proventriculus fails to form and the embryos fail to complete dorsal closure.

No difference is detected between the tilting of the nuclei of the circular visceral muscle (CVM) cells on the right and left sides of the presumptive first midgut constriction in late stage 15 and 16 homozygous embryos. This contrasts with wild-type embryos at these stages, where the nuclei of a subset of CVM cells in the right ventral region of the presumptive first chamber of the midgut become tilted compared with those in the left-ventral region.

38% of stage 17 pucGS16811/Df(3R)dsx10D embryos show inversion of the normal left-right asymmetry of the anterior midgut and 7% do not show any left-right asymmetry.

bsk1/+ significantly suppresses the defects in left-right asymmetry of the anterior midgut that are seen in homozygous pucGS16811 embryos.

Expression of bskDN.Scer\UAS under the control of Scer\GAL448Y significantly suppresses the defects in left-right asymmetry of the anterior midgut that are seen in homozygous pucGS16811 embryos.

Expression of bskDN.Scer\UAS under the control of Scer\GAL4Gap1-NP3392 significantly suppresses the defects in left-right asymmetry of the anterior midgut that are seen in homozygous pucGS16811 embryos. This suppression is stronger if the embryos are also carrying bsk1/+.

pucGS16811/+ significantly enhances the defects in left-right asymmetry of the anterior midgut which are seen in embryos expressing Rac1V12.Scer\UAS under the control of Scer\GAL4Gap1-NP3392. 20% of the double mutant embryos show inversion of the normal left-right asymmetry and 42% show no left-right asymmetry.

pucGS16811/+ has no effect on the defects in left-right asymmetry of the anterior midgut which are seen in embryos expressing Rho1V14.Scer\UAS under the control of Scer\GAL4Gap1-NP3392.

pucGS16811/+ has no effect on the defects in left-right asymmetry of the anterior midgut which are seen in embryos expressing Cdc42V12.Scer\UAS under the control of Scer\GAL4Gap1-NP3392.

    Comments

Expression of pucScer\UAS.cMa under the control of either Scer\GAL448Y or Scer\GAL4Gap1-NP3392 completely rescues the left-right asymmetry defects seen in the anterior midgut of pucGS16811 embryos. The defects in dorsal closure are not rescued.

The lack of left-right asymmetry in the tilting of the nuclei of the circular visceral muscle which is seen in late stage 15 and 16 pucGS16811 embryos is rescued by expression of pucScer\UAS.cMa under the control of Scer\GAL4Gap1-NP3392.

Expression of pucScer\UAS.cMa under the control of either Scer\GAL4dpp.PS or Scer\GAL4NP7020 does not rescue the left-right asymmetry defects seen in the anterior midgut of pucGS16811 embryos.

Expression of pucScer\UAS.cMa under the control of Scer\GAL4hs.PU using heat shock for 30 minutes at 37[o]C significantly rescues the left-right asymmetry defects seen in the anterior midgut of pucGS16811 embryos when the heat shock is applied at any time during 4.5 to 9.5 AEL.
