TrpA1W903 mutant larvae show a delayed nocifensive escape locomotion response compared to wild type flies. When gently touched with a 46[o]C probe many mutant flies fail to initiate escape behaviour within 10 seconds, compared to 3 seconds in wild type. Normal noxious heat responses are seen in TrpA1W903 mutant heterozygotes. TrpA1W903 mutants also show delayed mechanical nociception responses when stimulated with a 30mN von Frey fiber. However the response to gentle touch and gross motor functions both appear normal.

TrpA1W903/Df(3L)ED4415 mutant larvae show a delayed nocifensive escape locomotion response compared to wild type flies. When gently touched with a 46[o]C probe many mutant flies fail to initiate escape behaviour within 10 seconds, compared to 3 seconds in wild type. Only very mild noxious heat responses are seen in Df(3L)ED4415 mutant heterozygotes. TrpA1W903/Df(3L)ED4415 mutants also show delayed mechanical nociception responses when stimulated with a 30mN von Frey fiber. However the response to gentle touch and gross motor functions both appear normal.

Comments

Df(3L)ED4415 fails to complement the delayed nocifensive locomotion response seen in TrpA1W903. Df(3L)ED4415/+ larvae display only mild defects.

Expression of TrpA1+t21 rescues the thermal and mechanical nociception defects seen in TrpA1W903 mutant larvae.

Expression of TrpA1Scer\UAS.C.cZa under the control of Scer\GAL4ppk.1.9 rescues the thermal nociception phenotypes seen in TrpA1W903/Df(3L)ED4415 mutant larvae. In addition, expression of TrpA1Scer\UAS.C.cZa does not lower the behavioural threshold for thermal nociception. The mechanical nociception phenotypes seen in TrpA1W903/Df(3L)ED4415 mutants are not rescued.
