Detailed Description

Vps43B1 mutant clones in third instar larval eye discs undergo massive apoptosis. There is a reduction in the number of R8 photoreceptors compared to controls and R1-7 photoreceptor differentiation is disrupted.

Vps43B1 mutant clones in the wing disc survive poorly.

tub-GAL4

Vps43B1 mutant follicle cell clones are large in size, indicating normal survival of the mutant cells.

Ommatidia fail to differentiate in homozygous clones in the eye.

DroncI29 largely suppresses the increase in apoptosis seen in Vps43B1 mutant eye disc clones. The reduction in R8 photoreceptor number is also suppressed.

hepr75 does not suppress the photoreceptor differentiation defects or increased cell death seen in Vps43B1 mutant eye disc clones.

Expression of pucScer\UAS.cMa under the control of Scer\GAL4tub.PU does not suppress the photoreceptor differentiation defects or increased cell death seen in Vps43B1 mutant eye disc clones.

Expression of spis.Scer\UAS does not suppress the photoreceptor differentiation defects seen in Vps43B1 mutant clones, but induces ectopic photoreceptor differentiation in the wild type cells surrounding the clones.

Expression of btl::EgfrScer\UAS.T:λ\cI-DD does not suppress the photoreceptor differentiation defects seen in Vps43B1 mutant clones, but induces premature photoreceptor differentiation in the wild type cells surrounding the clones. The increase in cell death seen in Vps43B1 mutant clones is not suppressed by btl::EgfrScer\UAS.T:λ\cI-DD.

Expression of Ras85DV12.Scer\UAS suppresses the photoreceptor differentiation defects seen in Vps43B1 mutant clones. The increase in cell death seen in Vps43B1 mutant clones is also suppressed by Ras85DV12.Scer\UAS.

Vps43B1 suppresses the increase in photoreceptor number seen in shi7C7 mutant eye disc clones, with the double mutants showing the photoreceptor loss seen in Vps43B1 mutants alone. The same is seen in a DroncI29 mutant background.

Expression of shi1.Scer\UAS under the control of Scer\GAL4tub.PU does not suppress the photoreceptor loss seen in Vps43B1 mutant eye disc clones.

shiFL54 does not suppress the photoreceptor loss seen in Vps43B1 mutant eye disc clones (in which cell death has been prevented using DroncI29).

Comments

Expression of Vps4Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4tub.PU fully rescues the photoreceptor differentiation defects seen in Vps43B1 mutant clones.

98% of hemizygous Vps43B1 mutant males are rescued to viability by Vps4Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4tub.PU.
