Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 results in an increase in the average amplitude of the evoked excitatory junction potentials (eEJPs) as well as the quantal content at neuromuscular junctions of third instar larvae and the frequency distribution of miniature excitatory junction potential (mEJP) amplitude sizes is significantly different compared to controls, although the mean mEJP amplitude is unchanged.

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 results in an increased size of synaptic vesicles and a shift in frequency distribution toward larger vesicles in both type 1b and 1s of neuromuscular junction boutons in third instar larvae compared to controls.

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of either Scer\GAL4C164 or Scer\GAL4ple.PF results in a decrease in the distance travelled per unit of time by third instar larvae in a crawling assay compared to controls.

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control Scer\GAL4elav-C155 results in loss of dopaminergic neurons of the PPL1 cluster in the brain of adult flies.

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 results in defects in climbing behavior in adults of various ages (10, 20 and 30 day-old).

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 results in shortened lifespan of adult flies.

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of either Scer\GAL4C164 or Scer\GAL4ple.PF results in defects in locomotion and a significant decrease in the distance crawled by third instar larvae per unit of time compared to controls.

Ectopic expression of Hsap\SNCAScer\UAS.cKa under the control of either Scer\GAL4elav.PU or Scer\GAL4ple.PF results in loss of dopaminergic neurons from the PPL1 and PPM3 clusters in adult flies compared to controls.

When compared to controls, expression of Hsap\SNCAScer\UAS.cKa in serotonergic and dopaminergic neurons, under the control of Scer\GAL4Ddc.PL leads to a higher number of sleep episodes during the light phase but the average length of each sleep episode is not significantly affected. During dark phases, Hsap\SNCAScer\UAS.cKa expression leads to a reduction in the number of sleep episodes but an increase in the length of sleep episodes when compared to controlled flies. The overall level of activity is not affected.

Expression of Hsap\SNCAScer\UAS.cKa in serotonergic and dopaminergic neurons, under the control of Scer\GAL4Ddc.PL alters circadian locomotor activity. Anticipation behaviour is maintained in young (1-3 days after eclosion) and old (30 days after eclosion) flies expressing Hsap\SNCAScer\UAS.cKa. In young flies, Hsap\SNCAScer\UAS.cKa expression causes a phasing out of locomotion activity at the light-dark transition. This behaviour is lost in older flies.

Flies expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4Ddc.PL display wild-type circardian periodicity.

The number of dopaminergic neurons in the dorsolateral (DL) and dorsomedial (DM) clusters in the brain of flies expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav.PU is normal in both 2 and 29 day old adults.

25-30 day old flies expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav.PU have normal climbing ability.

Co-expression of Rab11Scer\UAS.T:Avic\GFP-EGFP suppresses increased eEJP amplitude, increased quantal content as well as increased synaptic vesicle size at boutons at the NMJs in third instar larvae expressing Hsap\SNCAScer\UAS.cKa driven by Scer\GAL4elav-C155

The defective crawling behavior (decrease in distance travelled per unit of time) characteristic for third instar larvae expressing Hsap\SNCAScer\UAS.cKa under the control of either Scer\GAL4C164 or Scer\GAL4ple.PF can be suppressed by co-expression of Rab11Scer\UAS.T:Avic\GFP-EGFP using either of the two drivers.

The defective crawling behavior (decrease in distance travelled per unit of time) characteristic for third instar larvae expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4C164 cannot be suppressed by co-expression of Rab11S25N.Scer\UAS.P\T.T:Avic\GFP-YFP.

The loss of dopaminergic PPL1 neurons in the adult brain characteristic for flies expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 can be reversed by co-expression of Rab11Scer\UAS.T:Avic\GFP-EGFP in 30 day-old flies.

The defects in climbing behavior seen in adult flies or various ages (10, 20 and 30 day-old) expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 can be suppressed by co-expression of Rab11Scer\UAS.T:Avic\GFP-EGFP.

The defects in climbing behavior seen in adult flies or various ages (10, 20 and 30 day-old) expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 are further worsened by co-expression of Rab11S25N.Scer\UAS.P\T.T:Avic\GFP-YFP.

The shortened lifespan characteristic for adult flies expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav-C155 can be reversed by co-expression of Rab11Scer\UAS.T:Avic\GFP-EGFP.

The locomotion defects characteristic for third instar larvae expressing Hsap\SNCAScer\UAS.cKa under the control of either Scer\GAL4C164 or Scer\GAL4ple.PF can be suppressed by co-expression of Rab8Scer\UAS.P\T.T:Avic\GFP-YFP : the decrease in the distance crawled by third instar larvae per unit of time is significantly ameliorated.

The reduction in the number of dopaminergic neurons from the PPL1 clusters characteristic for adult flies expressing Hsap\SNCAScer\UAS.cKa under the control of either Scer\GAL4elav.PU or Scer\GAL4ple.PF can be fully rescued by co-expression with Rab8Scer\UAS.P\T.T:Avic\GFP-YFP. The loss of dopaminergic neurons from the PPM3 cluster seen in adult flies expressing Hsap\SNCAScer\UAS.cKa under the control of Scer\GAL4elav.PU is partially restored by co-expression with Rab8Scer\UAS.P\T.T:Avic\GFP-YFP.
