Reference

Many homozygotes raised on sterilised corn-meal medium at 24[o]C and places on sterilised 20% agar plates at the pre-pupal stage can survive to the late pupal stage. Adult body formation is seen within the pupal cases, but none complete head eversion and no adults eclose. A similar phenotype is seen in animals derived from homozygous female germline clones.

The overall patterning of larval muscles is similar to wild type at 0 hours after puparium formation (APF) in homozygous larvae, although the shape of the muscles is slightly different from normal. At 10-25 hours APF, the morphology of the mutant myotubes is indistinct compared to wild-type controls. At later stages in the mutant animals, either the number of elongated myotubes is reduced and their positions are abnormal, or rounded myotubes appear instead of elongated myotubes, becoming randomly dispersed in the abdomen in some cases by 50 and 70 hours APF. Pupae that have failed to undergo head eversion also show defects in tendon cell distribution.

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4sr-md710 rescues head eversion in 56% of LamCLC58 pupae. Overall muscle morphology is restored to normal in these pupae.

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4how-24B rescues 89% of LamCLC58 animals to viability.
