Description

Imprecise excision of the P{PTT-GB} element, removing the P{PTT-GB} insertion and resulting in a 560bp deletion in the first LamC exon. This lesion is located within both the LamC and ttv transcription units (since LamC is nested within an intron of ttv). However, only LamC function is affected, since the "EX296" chromosome complements ttv00681b and fails to complement LamCEP2199.

LamCEX296/LamCEX265 transheterozygotes are lethal before adulthood and their third instar larval muscles 6 and 7 exhibit a significantly increased proportion of 'ghost' neuromuscular junctions, but not of abnormal nuclear lamina blebs, as compared to controls.

The expression of either LamCE174K.Scer\UAS or LamCR564C.Scer\UAS under the control of Scer\GAL4Mhc.PW in LamCEX296/LamCEX265 transheterozygotes leads to a strong increase in the number of nuclei exhibiting more than 10 abnormal lamina blebs in third instar larval muscles 6 and 7, as compared to controls.

Many homozygotes raised on sterilised corn-meal medium at 24[o]C and places on sterilised 20% agar plates at the pre-pupal stage can survive to the late pupal stage. Adult body formation is seen within the pupal cases, but none complete head eversion and no adults eclose.

LamCEX296 is early larval lethal when homozygous and late larval or early pupal lethal when hemizygous with Df(2R)trix.

Heterozygous LamCEX296 suppresses the position effect variegation of P{hsp26-pt-T}39C-72.

Heterozygous LamCEX296 suppresses the position effect variegation of T(2;3)SbV.

LamCEX296/Df(2R)trix animals show a developmental delay of about 2 days for all stages.

Comments

LamCEX296/LamCEX265 transheterozygotes exhibit lethality before adulthood, which is not rescued by the expression of LamCR564C.Scer\UAS under the control of Scer\GAL4Mhc.PW; LamCEX296/LamCEX265 transheterozygotes also exhibit an increased proportion of 'ghost' neuromuscular junctions in third instar larval muscles 6 and 7, which is rescued by the expression of LamCScer\UAS.cLa, but not of LamCE174K.Scer\UAS or LamCR564C.Scer\UAS, under the control of Scer\GAL4Mhc.PW.
