Larvae expressing Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP in class IV nociceptors under the control of Scer\GAL4ppk.1.9 exhibit robust nocifensive escape locomotion in response to blue-light illumination in 69% of animals.

Larvae expressing Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP under the control of Scer\GAL41003.3 and fed yeast paste containing all-trans retinal show gentle touch-like behavioural responses upon blue light illumination.

Expression of Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP under the control of Scer\GAL4tutl-GAL4 in the presence of all-trans retinal does not produce touch-like behavioural responses upon blue light illumination.

Third instar larvae expressing Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP under the control of Scer\GAL4109(2)80 and fed yeast paste containing all-trans retinal (the chromophore for Crei\ChR2) show a behavioural response to blue light pulses: the larvae simultaneously contract the muscles of all body segments and scrunch up like a compressed accordion. Rarely, the larvae first roll with a motor pattern that appears to be quite similar to nocifensive rolling behaviour before undergoing the accordion-like muscle contraction.

Third instar larvae expressing Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP under the control of either Scer\GAL4smid-C161 or Scer\GAL41003.3 and fed yeast paste containing all-trans retinal show a high frequency of accordion-like muscle contraction in response to blue light, but never show rolling behaviour.

Third instar larvae expressing Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP under the control of Scer\GAL4ppk.1.9 and fed yeast paste containing all-trans retinal show a high frequency of rolling behaviour in response to blue light which is qualitatively similar to nocifensive behaviour (although the initiation of the response is more rapid) , but never show an accordion-like muscle contraction.

Third instar larvae expressing Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP under the control of Scer\GAL42-21 and fed yeast paste containing all-trans retinal do not show either accordion-like muscle contraction or rolling behaviour in response to blue light.
