Expression of VhldsRNA.Scer\UAS.cMa under the control of Scer\GAL4tey-5053A results in an increase in the number of tracheal branches that terminate on ventrolateral body wall muscle 12 compared to controls.

Scer\GAL4btl.PS-mediated expression of strongly expressing VhldsRNA.Scer\UAS lines produces migration defects in the dorsal trunk and dorsal/ventral branch, and interruptions in the lateral trunk. Expression of milder lines leads o fewer migration and fusion defects per embryo and sinuous overgrowth of the primary and secondary branches. Knockdown also produces overgrown and intertwined secondary branches and secondary branch duplication.

During the third instar, Scer\GAL4btl.PS VhldsRNA.Scer\UAS larvae show increases in thick terminal branches and duplication of larval tertiary branches, and failure of lateral trunk fusion associated with cells terminating in multiple fine extensions.

Most Scer\GAL4btl.PS VhldsRNA.Scer\UAS animals develop through larval stages and die as pupae.

Scer\GAL4Act.PU-mediated expression of VhldsRNA.Scer\UAS leads to the appearance of ectopic tracheal placodes at the tp0 position and, at lower frequency, at the tp1 and tp11 positions.

Co-expression of agoΔF.Scer\UAS and VhldsRNA.Scer\UAS.cMa under the control of Scer\GAL4tey-5053A results in a synergistic increase in the number of tracheal branches that terminate on ventrolateral body wall muscle (VLM) 12 compared to larvae expressing either agoΔF.Scer\UAS or VhldsRNA.Scer\UAS.cMa alone under the control of Scer\GAL4tey-5053A. In addition to increasing the branching of the LF and LH terminal cells onto VLM12 compared to each single mutant, the double mutants show recruitment of ectopic branches from the LG lateral terminal cell onto VLM12 in approximately 10% of cases.

Co-expression of agoScer\UAS.cMa partially suppresses the phenotype of an increased number of tracheal branches terminating on VLM12 caused by expression of VhldsRNA.Scer\UAS.cMa under the control of Scer\GAL4tey-5053A.

simaKG07607/+ significantly reduces the penetrance of Scer\GAL4btl.PS VhldsRNA.Scer\UAS tracheal defects, while simaKG07607/simaKG07607 further suppresses these defects to background levels.

simaKG07607/+ completely suppresses the pupal lethality of Scer\GAL4btl.PS VhldsRNA.Scer\UAS.

btldev1/+ or bnl00857/+, but not trh10512/+, leads to a significant reduction in the penetrance of Scer\GAL4btl.PS VhldsRNA.Scer\UAS tracheal phenotypes.

ago3/+ enhances the tracheal phenotypes seen when Scer\GAL4btl.PS mediates expression of relatively weaker VhldsRNA.Scer\UAS lines, particularly enhancing the penetrance of migration defects.
