Scer\GAL4Crz.391 drives expression in 2 pairs of interneurons in the abdominal ganglion of males and in non-dimorphic neurons in the optic lobe and central brain. The interneurons in the abdominal ganglion are also labelled with Crz and fru proteins. The central brain neurons do not co-express fru.

Optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus with 650 nm (red) light) of Scer\GAL4Crz.391 cells induces ejaculation in males and results in a significant preference for an illuminated zone over non-illuminated areas, which is not seen in controls or in females; there is no effect on walking velocity, compared to controls; activation in combination with a paired odor induces a significant attractive olfactory memory that is not seen in controls; repeated activation over a few days results in a significant reduction in preference for ethanol-containing food, compared to controls.

Activation of Scer\GAL4Crz.391 neurons triggers ejaculation behavior; depending on the combination of UAS and light used, different numbers of flies show this behavior: around 20% with blue light activating Crei\ChR2Scer\UAS.cSa; 0% with blue light activation of Crei\ChR2H134R.Scer\UAS.2A.T:Avic\GFP-EYFP or Crei\ChR2C128T.Scer\UAS.T:Avic\GFP-YFP; 20% with red (0% with green light) activating Crei\ChR1[C1V1.T-T.Scer\UAS. T:Avic \GFP-EYFP]; 100% with red or green light activating Crei\ChR1ReaChR.Scer\UAS.T:Crei\ChR2,T:Avic\GFP-mCitrine.
