R68Q
Flies mutant for Ras85DR68Q exhibit a number of developmental defects that are characteristic of abnormally elevated RTK/RAS/MAPK signaling, including enhanced resistance to apoptosis, supernumerary R7 cells in the eye and ectopic wing vein formation.
Ras85DR68Q mutants have a reduced basal level of GTPase activity.
Ras85DR68Q mutants exhibit a statistically significant increase in the number of midline glia cells at stage 17 of embryogenesis, compared to controls.
Ras85DR68Q mutants exhibit supernumerary R7 cells in the developing ommatidia. Occasionally, ommatidia are found without outer photoreceptor cells, a phenotypic consequence of elevated MAPK signaling.
Ras85DR68Q flies show abnormalities in adult wing tissues. Homozygous Ras85DR68Q flies have an extra longitudinal 'veinlet' branching off the posterior crossvein. Additionally, an ectopic longitudinal vein is seen beneath the posterior crossvein and there is an ectopic crossvein found between the L4 and L5 wing veins near the hinge.

Ras85DR68Q is a suppressor | partially of hemocyte | larval stage phenotype of Scer\GAL4Hml.PG, hidUAS.cZa
Ras85DR68Q suppresses WGMR.PG induced cell death in a dosage-dependent manner.
Ras85DR68Q suppresses WAla3.GMR (which lacks 3 of the 5 predicted MAPK phosphorylation sites) induced cell death.
Ras85DR68Q fails to suppress WAla5.GMR (which lacks all of the 5 predicted MAPK phosphorylation sites) induced cell death.
Ras85DR68Q very mildly suppresses grimGMR.PC and rprGMR.PW induced cell death although this is suspected to be due to suppression of endogenous W activity and not grim or rpr activity.
A Ras85DR68Q background can partially suppress the hemocyte death seen upon expression of WScer\UAS.cZa under the control of Scer\GAL4Hml.PG. Hemocytes are clearly visible anteriorly in the lymph glands of 3rd instar larvae. Circulating hemocytes, however, appear to remain susceptible to W-induced cell death and are missing, even in the presence of two copies of Ras85DR68Q.
An Egfrf2 background does not affect the developmental and apoptotic defects found in Ras85DR68Q mutants.
A aosΔ7 background does not affect the developmental and apoptotic defects found in Ras85DR68Q mutants.
A rl10a background strongly ameliorates the developmental and apoptotic defects found in Ras85DR68Q mutants.
Ras85DR68Q is rescued by Ras85DN17.sev
Ras85DN17.sev expression in the eye abrogates the suppressive effects of Ras85DR68Q on WGMR.PG-induced apoptosis.
