Expression of Rab30dsRNA.Scer\UAS (with Dcr-2Scer\UAS.cDa) using either Scer\GAL4arm.PS or Scer\GAL4Act5C.PI gives a striking and penetrant dorsal closure phenotype: approximately 10–12% of prepared cuticles exhibit a severe dorsal closure phenotype, combined with total loss of anterior structures, and another 30–40% of cuticles showed a milder morphogenetic phenotype of partial failure in germband retraction, defects in head involution/specification and sometimes a dorsal pucker. An independent insertion of the same Rab30dsRNA.Scer\UAS transgene causes 45% of cuticles to exhibit only head involution defects, when expressed together with Dcr-2Scer\UAS.cDa by Scer\GAL4arm.PS, and a low percentage of morphological defects when expressed with Scer\GAL4Act5C.PI. Head involution defects are also produced (45% of cuticles) when Scer\GAL4en.PU is used to drive Rab30dsRNA.Scer\UAS with Dcr-2Scer\UAS.cDa.
A severe cleft thorax phenotype is produced by Scer\GAL4pnr-MD237-mediated expression of Rab30dsRNA.Scer\UAS with Dcr-2Scer\UAS.cDa, whereas a mild cleft thorax phenotype is produced by Scer\GAL4pnr-MD237-mediated expression of Rab30dsRNA.Scer\UAS without Dcr-2Scer\UAS.cDa.

The mild cleft thorax phenotype produced by Scer\GAL4pnr-MD237-mediated expression of Rab30dsRNA.Scer\UAS (without Dcr-2Scer\UAS.cDa) is enhanced in a background of hep1 heterozygosity or hemizygosity.
