Description

Insertion of a Mi{Hto-WP} element upstream of ct ( X:7 ,581,688 (+) -Release 6). The hostile takeover (hto) exon in the Mi{Hto-WP} element splices to the downstream ct exon 3. This results in a fusion protein being expressed under the control of Scer\UAS regulatory sequences in the Mi{Hto-WP} element. The fusion protein consists of 3 copies of the T:Zzzz\FLAG tag, the mCherry form of Disc\RFP and the coding sequence of the ct-RC isoform.

Reference

Expression of one copy of ctBRO under the control of Scer\GAL4GMR.PU produces flies that have pale/mottled and glazed (lacking clear separation of ommatidia) eyes.

Most flies expressing ctBRO under the control of Scer\GAL4ey.PU die during the pharate adult stage. Most of the pharate adult flies examined exhibit partial to full conversion of eye to antenna.

When ctBRO is expressed in the wing blade region under the control of Scer\GAL4Bx-MS1096 the distal part of the wing margin is strongly contracted and in the proximal part of the wing the alula is very reduced.

The legs of pupal stage flies expressing ctBRO under the control of Scer\GAL4ptc.PU show severe morphological changes.

Co-expression of PoxnBRB and ctBRO under the control of Scer\GAL4GMR.PU produces an eye phenotype that is qualitatively very similar to when either transgene is expressed alone. This phenotype is not suppressed upon co-expression of ctJF03304.

Expression of ctJF03304 substantially rescues the wing phenotypes seen when ctBRO is expressed under the control of Scer\GAL4Bx-MS1096. Only minor defects remain, such as a weak posterior margin.

Co-expression of ctBRO and ctJF03304 under the control of Scer\GAL4GMR.PU suppresses the eye phenotypes seen when either transgene is expressed alone. Only a slight reduction in pigmentation remains.
