Description

Re-integration of DNA into the attP site present in wgKO. The reintegrated sequence is as follows: 1. an FRT cassette containing the full-length, wild-type wg cDNA sequence (including 135 bp of 5'UTR and 1200bp of 3'UTR sequence), 2. 'Nrt-wg' (a membrane-tethered, Tag:HA-tagged form of wg in which Tag:M(Nrt) acts as the membrane tethering sequence), and 3. a loxP cassette containing a Disc\RFPmCherry.3xP3 marker. Nrt-wg is not expressed due to the presence of the upstream FRT cassette. In addition, a recombined attP/B site is present 5' to the reintegrated sequence.

Part of the wg first exon (including the ATG site and all the coding sequence of this exon) has been deleted. The deleted sequence has been replaced with the following reintegrated sequence: 1. an FRT cassette containing the wild-type wg coding region, 2. coding sequence for a membrane-tethered, Tag:HA-tagged form of wg (Tag:M(Nrt) acts as the membrane tethering sequence), and 3. a loxP cassette containing the Disc\RFPmCherry.3xP3 marker. The membrane-tethered, Tag:HA-tagged form of wg is not expressed due to the presence of the upstream FRT cassette. A recombined attP/B site is present 5' to the FRT cassette.

"wg{KO; WT[Body]; NRT-Wg[Discs]}" larvae expressing 'NRT-wg' (a membrane-tethered form of wg) only in imaginal discs (and wild-type wg in other tissues) have been generated by expressing FLPase in the imaginal discs of homozygous wgKO;FRT.wg.FRT.NRT-wg larvae (converting wgKO;FRT.wg.FRT.NRT-wg to wgKO;-FRT.NRT-wg in the discs). These larvae develop at the same rate as controls and eclose at near normal frequency when co-cultured with control siblings. The adult flies are 23% smaller than controls and the wing margin lacks occasional bristles.
