Description

A genomic fragment containing a 4kb region corresponding to CG14971 from 1.1kb upstream of the ATG to 1.9kb downstream of the stop codon, including the last exon of armi and 400bp downstream of the CG14971 stop codon (amplified from the BAC clone DS01105).

Co-expression of CG14971t4 does not further enhance the rescue of the Df(3L)armi-J mutant oogenesis defects by CycJ+t4.

Co-expression of CycJ+t4 further enhances the rescue of either Df(3L)armi-J or Df(3L)armi-J/Df(3L)Exel6095 mutant oogenesis defects by armi+t6.3. The hatching rate is improved from 30% to 73-76%.

A CycJnull/Df(3L)Exel6095 mutant background (expressing armi+t6.3 to restore armi[+] function) enhances the oogenesis defects seen in piwi06843/Df(2L)BSC145 mutant females. The frequency of compound egg chambers (multiple cysts in one follicular epithelium) is comparable to that seen in Df(3L)armi-J. Addition of CycJ+t4 decreases the frequency of compound egg chambers to the levels seen in the piwi mutant alone.

CycJnull/Df(3L)Exel6095 (expressing armi+t6.3 to restore armi[+] function) enhances the fertility defects seen in aubHN2/aubQC42 mutant females. The resulting flies lay no eggs and oocyte development is arrested, with egg chambers rarely advancing beyond stage 7-8. Compound egg chambers (multiple cysts in a single follicular epithelium) are seen, and these often include a disorganised layer of follicle cells almost separating two fully developed cysts. This phenotype can be fully suppressed by expression of CycJ+t4.

Comments

Expression of CycJ+t4 significantly rescues the oogenesis defects seen in homozygous Df(3L)armi-J females. Females are capable of producing fully developed stage 14 egg chambers and of laying eggs, albeit at a dramatically reduced rate compared to heterozygotes. There is also a dramatic decrease in the frequency of compound egg chambers (multiple cysts in one follicular epithelium) when CycJ+t4 is expressed in Df(3L)armi-J/Df(3L)Exel6095.
