Detailed Description
Statement

Females in which the eye is homozygous show electroretinogram defects.

The mean amplitude of the excitatory junctional current at the neuromuscular junction is not significantly different from that of controls in mutant larvae. However, the mean amplitude of the spontaneous miniature excitatory junctional current is significantly increased and quantal content is significantly decreased. FM 1-43 dye labelling indicates that synaptic vesicle recycling is reduced during stimulation. The readily releasable vesicle pool (RRP) is approximately 30% smaller in the mutant larvae compared to controls.

Additional Comments
Genetic Interactions
Statement
Reference

The electroretinogram defects seen in homozygous sky2 eyes are partially suppressed if the flies are also heterozygous for dor36.

The progressive retina neurodegeneration seen in flies containing homozygous sky2 eyes is suppressed if the flies are also heterozygous for dor36.

The increase in excitatory junctional current amplitude at the neuromuscular junction which is seen in sky1/sky2 larvae is suppressed if they are also heterozygous for dor36.

The increased synaptic vesicle trafficking to endosomes seen at the neuromuscular junction in sky1/sky2 larvae is not suppressed if the larvae are also heterozygous for dor36.

Comments

The increase in miniature excitatory junctional current amplitude and reduction in quantal content which is seen at the neuromuscular junction in dor36 larvae is rescued by expression of dor1-1002.Scer\UAS under the control of Scer\GAL4da.PU. The reduction in synaptic vesicle recycling is at least partially rescued.

Expression of dor1-1002.Scer\UAS under the control of Scer\GAL4VGlut.PU does not rescue the increase in miniature excitatory junctional current amplitude which is seen at the neuromuscular junction in dor36 larvae, while the amplitude of the evoked excitatory junctional current is significantly increased compared to the single mutant larvae. The reduction in synaptic vesicle recycling is at least partially rescued.
