Region containing the mir-965 hairpin is deleted via homologous recombination and replaced with a mini-white cassette. The deleted region was inferred from primer sequences.

mir-965KO1 mutant flies appear to be morphologically normal with the exception of defects in abdominal segmentation. The predominant defect is a dorsal gap in one or more abdominal segments, in some cases leading to segment fusion. Some of these flies also exhibit ectopic bristles associated with a polarity defect.
mir-965KO1/mir-965KO2 mutant flies appear to be morphologically normal with the exception of defects in abdominal segmentation. The predominant defect is a dorsal gap in one or more abdominal segments, in some cases leading to segment fusion. Some of these flies also exhibit ectopic bristles associated with a polarity defect.
mir-965KO1/mir-965KO2 mutant histoblasts undergo asynchronous division, resulting in the presence of cells of different sizes. In contrast to controls, a subset of the histoblasts undergo cell death. Subsequently the rate of histoblast nest expansion is slower in the mir-965KO1/mir-965KO2 mutants than in controls. In contrast to wild type, larval epidermal cells are still present in the nests.
Homozygous males and females show a significant increase in median lifespan compared to controls.
mir-965KO/Df(2L)ED19 males and females show a significant decrease in median lifespan compared to controls.

One copy of stgEY12388 partially suppresses the segment gap phenotype seen in the abdomens of mir-965KO1/mir-965KO2 mutant flies. The asynchronous histoblast division and reduction in histoblast migration rate are also suppressed.
One copy of stg4 partially suppresses the segment gap phenotype seen in the abdomens of mir-965KO1/mir-965KO2 mutant flies. The asynchronous histoblast division and reduction in histoblast migration rate are also suppressed.
One copy of wgSp-1 partially suppresses the segment gap phenotype seen in the abdomens of mir-965KO1/mir-965KO2 mutant flies.
One copy of wgl-12 does not significantly suppress the segment gap phenotype seen in the abdomens of mir-965KO1/mir-965KO2 mutant flies.
One copy each of stg4 and wgSp-1 partially suppresses the segment gap phenotype seen in the abdomens of mir-965KO1/mir-965KO2 mutant flies.
