Description

The R22B12 enhancer fragment is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain). A Hsp70 transcriptional terminator is present.

Inhibition (using shi^1.UAS at 31-33[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4^DBD.R22B12 and Hsap\RELA^AD.R30E11 in adults during training, consolidation and testing significantly reduces aversive 2 h associative olfactory memory, compared to controls, with no effect on innate odor or shock avoidance, compared to controls; inhibition (using shi^{ts1.IVS.p10.20xUAS} at 31-33[o]C) significantly reduces aversive 1 min associative visual memory, compared to controls, with no effect on naive shock avoidance.

Thermogenetic activation (using TrpA1^UAS.(B).cKa at 32[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4^DBD.R22B12 and Hsap\RELA^AD.R30E11 in combination with a paired odor significantly induces aversive olfactory 3 min memory, compared to controls.
