20 copies of the UAS site are fused upstream of the shi1 coding region (this encodes a temperature-sensitive dominant negative form of shi). A small intron derived from the Mhc gene has been inserted between the promoter and coding sequences. The 3' UTR (666bp) from the Autographa californica nucleopolyhedrovirus (AcNPV) p10 gene is present downstream of the coding sequence.

Blocking synaptic transmission in Tm5c neurons by expressing Ctet\TeTxLCTNT.Scer\UAS under the control of Scer\GAL4C1a.DBD.ort.T:Zzzz\ZipRREEL and Hsim\VP16AD.VGlut.OK371.T:Zzzz\ZipEERRL,T:SV40\nls2 at the non-permissive temperature significantly reduces the phototactic preference for ultraviolet light over green light seen in wild type and control flies.

Expression of shits1.IVS.p10.20xScer\UAS under the control of Scer\GAL4GMR48B04 converts the behavior of naive thirsty flies from water approach to water avoidance. This behavioral reversal is not evident at the permissive temperature.

Expression of shits1.IVS.p10.20xScer\UAS under the control of Scer\GAL4GMR48B04 has no effect on water avoidance in sated flies, suggesting that these flies perceive water normally and that output from R48B04 neurons is only required for water approach in thirsty flies.
