Description

The R58E02 enhancer fragment is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the p65(AD)::Zip+ hemidriver (component of a 'split driver' system, contains the p65 activation domain). A Hsp70 transcriptional terminator is present.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R48H11 and Hsap\RELAAD.R58E02, in combination with a Gal80[ts] transgene to restrict expression to the adult stage, does not affect food intake or climbing rate, compared to controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R48H11 and Hsap\RELAAD.R58E02 in adults does not affect food intake or climbing rate, compared to controls.

Optogenetic activation (using Zzzz\ChRChrimson.IVS.Syn21.10xUAS.tdTomato or Cnoc\ChR1Cs.IVS.20xUAS.Venus with 627 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R32D11 and Hsap\RELAAD.R58E02 in adult females results in a strong, slow depolarization of MBON-alpha1 (mushroom body output neuron 7), sufficient to elicit a spiking response.

Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 627 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R32D11 and Hsap\RELAAD.R58E02 in adult females, in combination with a paired odor, results in a short-term associative appetitive olfactory memory for this odor; preference is significantly greater when these neurons are not also activated during the testing period.

Acute thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R31F06 and Hsap\RELAAD.R58E02, in combination with a paired odor, in adults results in a significant increase in short-term associative aversive olfactory memory, compared to controls.

Inhibition (using shits.UAS.cUa at 33[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R31F06 and Hsap\RELAAD.R58E02 in adults significantly reduces short-term associative appetitive, but not aversive, olfactory memory, compared to controls, with no effect on sugar preference.

Inhibition (using shi1.UAS at 32[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R37E10 and Hsap\RELAAD.R58E02 in adults during, or immediately after, training, but not during, or immediately before, testing, significantly impairs long-term appetitive olfactory memory, compared to controls; inhibition does not affect short-term memory.

Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 617 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R93G08 and Hsap\RELAAD.R58E02, the split-GAL4 pair of Scer\GAL4DBD.R18D09 and Hsap\RELAAD.R58E02, or the split-GAL4 pair of Scer\GAL4DBD.R22E04 and Hsap\RELAAD.R58E02, in adults results in a significant attraction to the illuminated area; activation of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R93A09 and Hsap\RELAAD.R58E02 results in a non-significant attraction to the illuminated area, no attraction is observed in controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R37E10 and Hsap\RELAAD.R58E02 in adults, in combination with a paired odor, results in a significant increase in long-term associative attractive olfactory memory, compared to controls.

Inhibition (using shits1.IVS.p10.20xUAS at 33[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R37E10 and Hsap\RELAAD.R58E02 in adults during training significantly reduces long-term, but not short-term, associative appetitive (sugar) olfactory memory, compared to controls.

FlyBase curator comment: According to correspondence with the authors, some split combinations listed in the methods of the paper are incorrect; the correct combinations (MB002B=R12C11-AD+R14C08-DBD, MB316B=R58E02-AD+R93G08-DBD, MB056B=R76F05-AD+R80G12-DBD, MB047B=R58E02-AD+R93A09-DBD) are listed on FlyLight so these are the ones that have been associated with the paper.
