Description

2.5kb deletion that removes most of the WRNexo coding sequence (deletion extends from 426bp upstream of the 5'UTR to 17bp upstream of the 3'UTR). Generated by imprecise excision of P{EP}G16048. Expression of the upstream Nup43 gene is not affected.

Comment:

Inferred boundaries of a 2.5 kb deletion resulting from the imprecise excision of P{EP}G16048, which is reported to delete sequences from 426 bp upstream of the 5'-UTR to 17 bp upstream of the 3'-UTR. The coordinates used for the ends of the UTRs were not defined.

Detailed Description
Statement
Reference

WRNexoΔ/WRNexoΔ flies have normal survival rates in response to camptothecin or topotecan treatment and are not sensitive to bleomycin.

WRNexoΔ/WRNexoΔ mutants have significantly reduced hatching frequency and embryos have more nuclear division defects (increased number of anaphase bridges and gaps between nuclei) and significantly increased double-strand breaks compared to wild type.

Larval exposure to hydroxyurea decreases (dose-dependently) WRNexoΔ/WRNexoΔ or WRNexoΔ/Df(3R)Exel6178 adult survival rates.

WRNexoΔ/WRNexoD229V mutants do not have reduced hatching frequency or hydroxyurea sensitivity (except hatching frequency is significantly reduced when embryos are laid by WRNexoΔ females mated with WRNexoD229V/WRNexoΔ males).

Genetic Interactions

spn-A3/spn-A3 enhances hydroxyurea sensitivity (decreases adult survival) in WRNexoΔ/WRNexoΔ mutants. BlmN1/BlmN1 WRNexoΔ/WRNexoΔ double mutants have similar hydroxyurea sensitivity to either single mutant.

Synthetic lethality is observed in double mutants: WRNexoΔ/WRNexoΔ mus81Nhe/mus81Nhe survive to adulthood but homozygotes eclose at lower frequencies than expected and show poor survival; WRNexoΔ/WRNexoΔ mus312D1/mus312D1 survive to pharate adult stage; WRNexoΔ/WRNexoΔ GenZ4325/GenZ4325 survive to pupal stage. WRNexoD229V/WRNexoD229V mus81Nhe/mus81Nhe double mutants survive to adulthood and are healthy.
