Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change.
Compared to wild type (pupate around day 5, eclose around day 10), Tcs31A13/Df(3L)ED220 exhibit an extended larval period prior to lethality (viability: 100% at day 8, 40% at day 16) and have melanotic masses ('black spots': 95% at day 8, 100% at day 16); larvae are larger than normal.
Compared to control Tcs31A13/+ heterozygotic third instar larval hemolymph (made up of primarily plasmatocytes and more rare lamellocytes), Tcs31A13/Df(3L)ED220 hemolymph from 10 day old larvae shows increased blood cell density and massive overrepresentation of lamellocytes, along with circulating aggregates resembling lymph gland fragments and tumors packed with lamellocytes. Tcs31A13/Df(3L)ED220 larval lymph glands show abundant lamellocyte maturation, are enlarged and the anterior lobe is detached or detaching from the dorsal vessel; second instar enlarged lymph glands show signs of increased cell division.
Most Tcs31A13/Df(3L)ED220 third instar larvae are missing imaginal discs and have a slightly smaller CNS; optic lobes are reduced in size, while the ventral nerve cord is less affected. Tcs31A13/Tcs31A13 mutant clones are not recovered in wing imaginal discs.
Tcs31A13/Df(3L)ED220 have significantly smaller salivary glands and salivary gland cell size is slightly but significantly reduced compared to wild type larvae; size of body wall musculature (length of ventral longitudinal muscles 1 and 2) is not significantly affected.
Df(3L)ED220/Tcs31A13 is rescued by Tcs3GFP(S65T)
Df(3L)ED220/Tcs31A13 is rescued by Tcs3+tLa
Tcs3T:Avic\GFP-S65T or Tcs3+tLa rescues Tcs31A13/Df(3L)ED220 to adult viability and no melanotic masses are seen in rescued animals.
