Electroretinograms (ERGs) recorded from wcyA/wcyA mutant eye clones show significantly reduced amplitudes and off transients compared to controls; at day 1, mutant clones show disruption of ommatidial units, with abnormally fused or split rhabdomeres and loss of photoreceptor cells and mutant clones also show show age-dependent (1 day old versus 36 days old) degeneration of internal eye structures (defects are more severe than day 1 and numerous photoreceptors are lost, leaving vacuoles). One day old mutant retina and lamina clones show an increase in autophagosomes, amphisomes, autolysosomes and lysosomes compared to controls.

wcyA/wcyA mutant clones in starved early third instar larval fat bodies show decreased numbers of autophagosomes and lysosomes compared to surrounding control cells. Fed wcyA/wcyA mutant clones also show an increase in numbers of autophagosomes and lysosomes in fat body cells of second and third instar larvae compared to controls. Autophagosome-lysosome fusion occurs in wcyA/wcyA second instar larval fat bodies.

wcyA/wcyA full eye clones are reduced in size compared to controls. wcyA/wcyA mutants exhibit slow growth and developmental delay.

wcyA/wcyA mutants in egg chamber follicle cells show reduced clone and cell size.

Expression of Atg1TRiP.cUa driven by Scer\GAL4cg.PU suppresses increases in autophagosomes seen in wcyA/wcyA fat body third instar larval clones.

Expression (at 18[o]C for low level expression) of TorWT.Scer\UAS.T:Zzzz\FLAG driven in the fat body by Scer\GAL4cg.PU suppresses the autophagy phenotype of wcyA/wcyA clones; expression driven in the retina by Scer\GAL4ninaE.PU partially significantly suppresses autophagy phenotypes (increased number of autophagosomes, amphisomes, autolysosomes and lysosomes) seen in wcyA/wcyA clones, along with improving synaptic transmission (significantly increasing off transient amplitude).

Complementation and Rescue Data
Comments

Expression of wcyScer\UAS.cDa or wcyScer\UAS.T:Zzzz\FLAG driven by Scer\GAL4Ubi.PU or wcy+tGR, wcyT:Zzzz\FLAG,T:Ivir\HA1 or wcyT:Avic\GFP-EGFP rescues lethality in wcyA/wcyA mutants. wcy+tGR rescues retinal morphology and electroretinogram defects in wcyA/wcyA mutants. wcy+tGR rescues starvation induced autophagy defects in wcyA/wcyA third instar larval fat body clones. wcy+tGR, wcyT:Zzzz\FLAG,T:Ivir\HA1 or wcyT:Avic\GFP-EGFP rescues the programmed autophagy phenotype in wcyA/wcyA third instar larval fat body clones.
