Description

Deletion that spans the translation initiation codon common to the A and B isoforms of TrpA1, preventing expression of these two isoforms. Sequence encoding a GAL4 driver and a w+* marker have been inserted in place of the deleted sequence. The GAL4 driver acts as a reporter of expression of the TrpA1 A and B isoforms.

Sequence encoding a GAL4 driver has been inserted at the site of the normal TrpA1 translation initiation codon, together with the simultaneous deletion of a 185 bp region encoding the N-terminal 61 residues of TrpA1.

Inferred boundaries of a 185bp deletion generated by ends-out homologous recombination that spans the ATG codon of TrpA1 and removes the N-terminal 61 amino acid residues. A sequence encoding a GAL4 driver has been inserted at the site. The deletion appears to remove the ATG and amino terminus of the TrpA1 -PI and -PJ isoforms and not the TrpA1 -PG, -PH, and -PK isoforms but it was not specified.

TrpA1-AB-G4 homozygous, but not heterozygous, mutant larvae have severely reduced rolling (escape) behavior in response to noxious temperature increases, compared to controls.

TrpA1GAL4 flies show lack of uracil-dependent defecation.

TrpA1AB-G4 homozygous mutant third instar larvae show impaired temperature selection and unlike controls display no strong preference for cooler temperatures (18[o]C) in a temperature gradient.

At a constant temperature of 25[o]C, the evening anticipation shown by mutant flies under 12 hour light/12 hour dark conditions is similar to that seen in wild -type flies. The mutant flies also show morning anticipation, although it appears to be slightly reduced. Free-running flies under constant darkness conditions retain morning and evening peaks and the periodicities are similar to wild type. Morning anticipation under constant darkness also appears to be reduced in the mutant flies under constant darkness.

Mutant flies show differences in activity compared to wild type under thermophase/cryophase (TC) conditions (flies entrained initially under 12 hour light/12 hour dark conditions for 4 days at 18[o]C, followed by 7 days of thermophase (18 hours at 29[o]C) and cryophase (6 hours at 18[o]C) cycles in constant darkness). During the first TC cycle, the mutant flies show double peaks, with the second peak approximately coinciding with the original morning peak established during the light-dark entrainment. The morning peaks during the TC cycles are reduced. The evening activity shown by the mutant flies initiates slightly earlier than wild type and is broader. The mutants require an additional day during the TC cycling compared to wild-type to establish a constant period between the evening activity peaks.

TrpA1GAL4 flies display a reduced aversion to aristolochic acid compared to wild type.

In contrast to wild type, TrpA1GAL4 flies have virtually no aristolochic acid-induced action potentials in tip recordings of sensillum s6.
