In a split-GAL4 system, Hsap\RELAAD.R24C08 drives expression in antennal grooming brain interneuron 2, the adult central complex and the posterior part of the brain when combined with Scer\GAL4DBD.R26B12. Antennal grooming brain interneuron 2 were seen in close proximity to Ecol\lexAGMR27H08 expressing zone C/E Johnston organ neurons. Positive GRASP (GFP reconstitution across synaptic partners) signals were seen between C/E Johnston organ neurons and antennal grooming brain interneuron 2 using these drivers.
Thermogenetic activation (using TrpA1Scer\UAS.(B).cKa at 30-31[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R26B12 and Hsap\RELAAD.R24C08 results in a significant increase of antennal grooming behavior compared to control flies; there is a further significant increase in antennal grooming behavior following antennal amputation.
Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with red light activation) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R26B12 and Hsap\RELAAD.R24C08 induces antennal grooming, which persists for tens of seconds beyond red light cessation.

Inhibition of synaptic release (using Ctet\TeTxLCTNT.Scer\UAS) in neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R26B12 and Hsap\RELAAD.R24C08 significantly partially suppresses the increase in antennal grooming seen in flies with thermogenetic activation (using TrpA113xlexAop.JFRC26 at 30-31[o]C) of Ecol\lexAGMR27H08 neurons.
