UAS regulatory sequences drive expression of the AdamTS-A-RB isoform (derived from the GH16393 cDNA clone).
Expression of AdamTS-AUAS.RB under the control of Scer\GAL4elav.PU (combined with Scer\GAL80repo.PL) has no effect on larval brain morphology, however expression controlled by Scer\GAL4repo results in an elongated nerve cord and flattened brain lobes. Similar morphological defects are also observed upon Scer\GAL4repo-driven expression of AdamTS-AUAS.RB in the AdamTS-Arnwy1/AdamTS-Arnwy2 mutant larvae.
AdamTS-AUAS.RB, Scer\GAL4repo has abnormal neuroanatomy | third instar larval stage phenotype, non-suppressible by trolG0271/trolG0271
AdamTS-AUAS.RB, Scer\GAL4repo has abnormal neuroanatomy | third instar larval stage phenotype, non-suppressible by trolsd/trolsd
AdamTS-AUAS.RB/Scer\GAL4repo is a non-suppressor of abnormal developmental rate | larval stage phenotype of trolsd
AdamTS-AUAS.RB, Scer\GAL4repo has larval ventral nerve cord | third instar larval stage phenotype, non-suppressible by trolG0271/trolG0271
AdamTS-AUAS.RB, Scer\GAL4repo has larval ventral nerve cord | third instar larval stage phenotype, non-suppressible by trolsd/trolsd
The abnormal elongation of ventral nerve cord observed in third instar larvae expressing AdamTS-AUAS.RB under the control of Scer\GAL4repo is not prevented by homozygosity of trolG0271 or trolsd. However, expression of AdamTS-AUAS.RB under the control of Scer\GAL4repo does not suppress the larval growth defects characteristic for trolsd mutants and the larvae still arrest their development at second instar stage.
Scer\GAL4elav.PU, AdamTS-AUAS.RB, Scer\GAL80repo.PL fails to rescue AdamTS-Arnwy2/AdamTS-Arnwy1
Scer\GAL4NP577/AdamTS-AUAS.RB fails to rescue AdamTS-Arnwy2/AdamTS-Arnwy1
Expression of AdamTS-AUAS.RB driven by Scer\GAL4elav.PU fails to rescue the neural invasion phenotype (neural cells from the brain invade adjacent peripheral structures such as imaginal discs) characteristic for AdamTS-Arnwy1/AdamTS-Arnwy2 mutant larvae when combined with Scer\GAL80repo.PL, but the phenotype is rescued if the driver is used alone. Scer\GAL4repo- or Scer\GAL4Gli-rL82-driven expression also rescues the invasion phenotype but replaces it with severe morphological anomalies: an elongated nerve cord and flattened brain lobes. Expression of AdamTS-AUAS.RB controlled by Scer\GAL4NP577 does not rescue the neural invasion.
