ScerGAL4NP1562 drives expression in three clusters of 19-22 neurons in total in the adult brain; neurons that are in and around the antennal mechanosensory and motor center including the sweet gustatory projection neurons, and two clusters of neurons in and around the dorsal anterior and ventral parts of the adult gnathal ganglion.
Inhibition (using Ctet\TeTxLCTNT.UAS or Hsap\KCNJ2UAS.EGFP) of Scer\GAL4NP1562 neurons, in combination with a Gal80[ts] transgene to restrict expression to the adult stage, significantly reduces sucrose feeding across a range of concentrations, compared to controls; proboscis extension responses to tarsal sucrose stimulation are also significantly reduced when Ctet\TeTxLCTNT.UAS is used.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 32[o]C) of Scer\GAL4NP1562 neurons induces a high frequency of proboscis extension, compared to controls; activation (at 30[o]C) does not affect sucrose feeding, compared to controls.
Adult males in which clones (generated using hs-FLP during larval stage) of Scer\GAL4NP1562 neurons are thermogenetically activated (using TrpA1UAS.(B).cKa at 32[o]C) always have an induction of proboscis extension, compared to controls, when the sweet gustatory projection neurons (cluster I) are activated; this is not the case for other types of Scer\GAL4NP1562 neuron.
