7 days old SH3PX1d1 homozygotes display severely increased numbers of progenitor cells (Mira-positive intestinal stem cells and enteroblasts) and of mitotic cells (phospho-H3 staining) in the adult midgut, as compared to heterozygous and wild-type controls. Likewise, SH3PX1d1/SH3PX1HK62b transheterozygotes also show a significant increase in the number of mitotic cells (phospho-H3 staining), as compared to SH3PX1d1 heterozygous controls.
SH3PX1d1 homozygous midgut clones generated during adulthood are significantly larger than control clones, but present similar frequencies of enterocytes (Pdm1-positive) and enteroendocrine cells (Prospero-positive), as compared to controls. The clonal cells fail to show the expected starvation-induced autophagosome formation (identified as Atg8-positive puncta) normally observed in controls.
