iPLA2-VIAnull homozygous adults show a severe and progressive decrease in climbing activity, disturbance in sleep, namely a significant increase in day time sleep and a significant and progressive decrease in night time sleep, and heat sensitivity. Their brains show vacuolization, a progressive decrease in the number of DA neurons (PPL1, PPM1/2 and PPM3), and ER stress. In light microscopy of DA neurons and indirect flight muscles, mitochondrial morphology appears normal. In TEM of the adult brain, there are aberrant crista dilation, multilamellar bodies and highly electron-dense structures.

iPLA2-VIAnull homozygous third instar larval show poorly-developed salivary glands with extended ER and signs of ER stress, as compared to controls. Their neuromuscular junctions show more and smaller synaptic vesicles, despite of no changes in docked vesicle number, bouton number and size or activation zone number. Neuromuscular neurotransmission shows a significant decrease in mEJP amplitude at 22[o]C and 30[o]C, but no major changes in EJP amplitude and quantal content, as compared to controls; however, PPR is reduced at 30[o]C, but not at 22[o]C, as compared to controls.

The extended ER observed in the salivary glands of iPLA2-VIAnull third instar larvae is suppressed by the Scer\GAL4elav-C155-driven expression of Hsap\C19orf12UAS.cMa or Hsap\PLA2G6UAS.cMa, even leading to a decrease in ER area.
