Subject: Revised FB entry for Tpl
--========================_64989128==_
Dear Colleagues,
I have attached a revised version of the FB entry for Tpl. The revisions
consist of correction of errors that date back to the DIS version of
Lindsley and Zimm, as well as corrections from new data recently published
(Dorer et al, Dec. 1995, Genetics 141(3): 1037-1042. The revision was
constructed by downloading the current entry as text, and then editing it.
I have noted lines that need to be added or changed from the current entry
with a \*, lines that need to be deleted with a >, and lines that consist of
my comments (not to be included in the entry, but to explain to you why a
change is necessary) with a ~. If you have any questions or if my mailer
does not attach the file properly to this message please let me know.
Sincerely,
Alan Christensen

Gene symbol Tpl
\*Full name Triplo-lethal
=46lyBase ID number FBgn0003739 Graphic map
Synonym(s) tpl
Date 13 Oct 95
\*Genetic map position very close to Ki because deletions of Tpl
\* also delete Ki (Roehrdanz and Lucchesi, 1980),
\* and because no deletions of Ki were obtained
\* presumably due to the close proximity of Tpl
\* (Kaufman, 1978)
Cytological map position
Located in 83D5-E1 because included in Df(3R)Tpl4 =3D
   Df(3R)83D4-5;83E1-2 and Df(3R)Tpl7 =3D Df(3R)83D4-5;83E1-2 (Keppy
   and Denell, 1979).
Revised cyto. map posn. 83E1--83E2
Phenotypic information
Unique dosage-sensitive locus at 83D-E; first detected by
segmental aneuploidy, using the Y-autosome translocations
T(Y;3)L132 and Tp(Y;3)A109 (Lindsley, Sandler, Baker, Carpenter,
Denell, Hall, Jacobs, Miklos, Davis, Gethmann, Hardy, Hessler,
Miller, Nozawa, Parry and Gould-Somero, 1972). Lethal when
present in either one (Tpl/Df) or three doses (Tpl/Dp) in an
otherwise diploid individual. These individuals do not survive
to the adult stage, but a few larvae with three doses of 83D-E
develop to the third instar. The surviving larvae are also
hyperploid for the X chromosome (as in 3X;2A metafemales); they
can be produced in genotypes duplicated for 7C and 7D-E
\* (Roehrdanz and Lucchesi, 1979; Roehrdanz and Lucchesi 1981). This
\* interaction is due to the Isis locus (Dorer et al., 1993). Flies
\* with a deficiency for 83D-E in one chromosome 3 and a duplication for
the region in the other (two doses in all) are viable (Denell,
\* 1976). Crosses of these Df/Dp flies to wild type
\* fail to produce viable adults (Keppy and Denell, 1979).
\* When wild-type flies
\* are treated
with EMS or gamma-rays (Keppy and Denell, 1979; Roehrdanz and
\* Lucchesi, 1980), new deficiencies of Tpl and mutations in the tightly li=
nked
\* Su(Tpl) locus are obtained (Dorer et al., 1995). No function in measurin=
g
\* the X/A ratio has been observed in Tpl; Tpl-aneuploids are not rescued
by mutations in
\* the sex-determining genes Sxl and da (Christensen and
   Lucchesi, 1988).
~Delete the entire paragraph of Miscellaneous info, as it is incorrect
>Miscellaneous info...
Data from ref. FBrf0051978
  Phenotypic information
\* Hybrid dysgenesis has been used to mutate the Tpl locus, but no amorph=
ic
\* alleles were generated, suggesting Tpl may be a complex locus.
~Delete this sentence because it really refers to Su(Tpl)
> A higher mutation rate with hybrid dysgenesis than with
> radiation or chemicals suggests a peculiar genetic
> organization of Tpl.
Data from ref. FBrf0058587
  Phenotypic information
\* Increased dosage of the Is locus suppresses the triplo-lethal, but
\* not the haplo-lethal phenotype of Tpl. Altered dosage of Is in
the presence of two copies of Tpl has no obvious effects. Is
probably does not directly repress Tpl expression, but rather
acts downstream of the Tpl triplo-lethal phenotype.
Data from ref. FBrf0066351
  Phenotypic information
Flies with three or one dose of Tpl die as late embryos or early
first instar larvae. Lethality can be rescued by dosage of the
\* Is locus, or mutations of the Su(Tpl) locus

  Phenotypic information
Mutations at Su(Tpl) block the triplo-lethal effect of Tpl.
Several alleles previously thought to be hypomorphic mutations
of Tpl are dominant mutations of the tightly linked Su(Tpl).
\* Su(Tpl) mutations are also recessive lethal.
