P{10XUAS(loxP.stop)GFP.Myr} and P{10XUAS(FRT.stop)GFP.Myr} are similar to the recombinase tester constructs described in Nern et al. (FBrf0214794). A stop codon-bearing cassette is flanked by loxP or FRT sites so that a GFP variant (containing F64L and S65T amino acid substitutions and which has been optimized for Drosophila codon usage) with a myristoylation tag is expressed in the presence of Cre or FLP recombinase and an appropriate GAL4 driver.
P{3XUAS-RSR.PEST}is similar to the recombinase constructs described in Nern et al. R recombinase tagged with a PEST sequence is expressed under the control of three UAS copies.
P{UAS-B2R.PEST}, P{UAS-B3R.PEST}, P{UAS-KDR.PEST} and P{UAS-RSR.PEST} were constructed by cloning the PEST-tagged B2, B3, KD and R recombinase sequences described in Nern et al. into a vector similar to P{1XUAS-IVS- mCD8::GFP } (pJFRC3; FBtp0056314) in which the Hsp70 promoter had been replaced by a Drosophila synthetic core promoter (DSCP; FBrf0205679). P{UAS-cre.PEST} is a similar construct with PEST-tagged Cre.
