P{UAS-ryA} expresses the same zinc finger nuclease directed against the ry gene that is encoded by P{hs-ryA} (FBtp0021952) under UAS control.
P{UAS-ryB} expresses the same zinc finger nuclease directed against the ry gene that is encoded by P{hs-ryB} (FBtp0021953) under UAS control.
P{hs-yA.P501T.I538K} expresses the same zinc finger nuclease encoded by P{hs-yA} (FBtp0016058) except for substitution of sequences encoding Threonine for Proline at residue 501 and Lysine for Isoleucine at residue 538. The P501T substitution corrects a missense mutation to give the wild type FokI sequence and the I538K substitution prevents homodimerization of the zinc finger nuclease. The construct was generated in a Casper-based vector.
P{hs-yB.Q486E.P501T} expresses the same zinc finger nuclease encoded by P{hs-yB} (FBtp0016057) except for substitution of sequences encoding Threonine for Proline at residue 501 and Glutamic acid for Glutamine for at residue 486. As above, the P501T substitution corrects a missense mutation to give the wild type FokI sequence. The Q486E substitution prevents homodimerization of the zinc finger nuclease. The construct was generated in a Casper-based vector.
P{hs-ryA}1C, P{hs-yA}20b, P{hs-yB}7a, P{hs-yA.P501T.I538K}2C, P{UAS-ryA}14C and P{UAS-ryB}1B are second chromosome insertions.
P{yM.donor}3, P{hs-ryB}2C and P{hs-yB.Q486E.P501T}2C are third chromosome insertions.
