FLP-mediated recombination between the FRT sites present in TI{GFP}E(spl)mδ-HLHGFP.NLS.w+ and TI{mCherry}E(spl)m8-HLHmCherry has deleted the intervening genomic sequence, resulting in a deletion that extends from E(spl)mδ-HLH to E(spl)m8-HLH. The E(spl)mδ-HLH coding sequence is deleted from the ATG codon onwards, while the 5' end of the E(spl)m8-HLH locus is deleted (up to and including the ATG codon). Sequence encoding GFP, Tag:MYC and Tag:NLS(Unk), followed by SV40 termination sequences is inserted at the site of the deletion (represented by TI{GFP}Df-E(spl)mδ-m8). A single FRT site is present downstream of the SV40 termination sequences.
