An elav promoter regulates expression of the major isoform (SC3ORF) of ewg.

Construct: ewg cDNA, SC3, with 27 nucleotides of 5' untranslated leader and 600 nucleotides of 3' trailer under the control of the elav promoter.

Flies expressing ewgelav.NS in a ewg2 mutant background are viable but display loss of indirect flight muscles. The usual six dorsal longitudinal (DLMs) and seven dorsal ventral muscles (DVMs) on each side of the midline are all lost to be replaced by two large misshapen muscles. The photoreceptors appear normal.

Myoblasts are present, migrate normally and get distributed over the persistent thoracic larval muscles in animals expressing ewgelav.NS in an ewg11 background, but splitting of the persistent larval muscles does not occur in some genetic backgrounds. Fusion of the imaginal myoblasts with the larval muscles occurs normally, but the wild-type progressive alignment of fused myoblast nuclei into longitudinal columns does not occur in these animals.

Expression can rescue embryonic lethality of ewg mutants but not the adult muscle defects.

Hemizygous ewgP1 suppresses the apoptosis seen in ApcQ8 mutant photoreceptors. This apoptosis can be restored by expressing ewgelav.NS.

elavts1/elav5 animals eclose as adults when they are rescued by ewgelav.NS (providing that embryogenesis occurs at the permissive temperature for the elavts1 allele). However, the rescued adults show motor defects and are flightless.

The reduction in bouton number at the neuromuscular junction which is seen in elavts1/elav5 animals reared at the restrictive temperature during larval life is rescued by ewgelav.NS.
