Expression of RafK497M.Scer\UAS driven by Scer\GAL4dpp.PU does not lead to tissue overgrowth in eye-antennal imaginal or wing discs.

Expression of phlK497M.Scer\UAS under the control of Scer\GAL4btl.PS results tracheal branching defects in embryos.

Expression of phlK497M.Scer\UAS under the control of Scer\GAL4amn-c651, results in pupae and adults that are larger than wild-type controls. The wing epidermis of these flies is increased, suggesting that most of this increased size results from increased cell size rather than cell number. Expression of this transgene under the control of Scer\GAL4amn-X8 causes an increase in pupal length. Flies that express one copy of phlK497M.Scer\UAS, under the control of one copy of either Scer\GAL4amn-c651 or Scer\GAL4amn-X8, pupariate about 36 hours after wild-type controls, which results mostly from a prolonged third instar stage. Expression of phlK497M.Scer\UAS under the control of Scer\GAL4amn-c651 has no effect on prothoracic gland cell size.

When phlK497M.Scer\UAS is driven by Scer\GAL4dpp.blk1 no effect is seen on wing disc growth.

phlK497M.Scer\UAS; Scer\GAL4prd.RG1 embryos do not have obvious cuticle phenotypes.

phlK497M.Scer\UAS; Scer\GAL4Ubx-lac1-Gal4 adults exhibit wing duplication in the posterior compartment and partial loss of notum and/or hinge. Occasionally pharate adults die within the pupal case, with severe loss of notum tissue. In larvae of this genotype, the wing discs show altered morphology, particularly overgrowth in the posterior hinge and mesonotum. Marker analysis indicates formation a new wing pouch complete with the dorso-ventral boundary, and that pattern duplication is associated with loss of notum, confirming notum/hinge-to-wing transformations.

When expression is driven by Scer\GAL4C-765, anterior supraalar bristle and posterior supraalar bristle bristles are usually eliminated and approximately 45% of heminota show an extra dorsocentral bristle. When expression is driven by Scer\GAL4sca-C253, only the anterior supraalar bristle is eliminated and only occasionally duplicates a dorsocentral bristle. When expression is driven by Scer\GAL4pnr-MD237, dorsocentral bristles are absent. Expression driven by Scer\GAL4179b causes lethality and the few pharate adults recovered lack 50-80% of notum macrochaetae, though in half of the cases the socket of the missing bristle was present.

When expression is driven by Scer\GAL4twi.PB the number of HVM cells is reduced.

phlK497M.Scer\UAS, when driven by Scer\GAL4en-e16E leads to a posterior to anterior transformation of the wing margin. Overexpression of phlK497M.Scer\UAS in clones driven by Scer\GAL4Act5C.PI and induced at 36+-12h after egg laying can produce mirror image duplications of the wing and notum to wing transformations.

When expression is driven by Scer\GAL4en-e16E the wing is reduced in size and veins L4 and L5 are absent.

Expression of phlK497M.Scer\UAS under the control of Scer\GAL4sca-537.4 results in flies that lack pieces of wing veins L3 and L4. Ectopic veins are also seen.

Hyperplasia in adult Malpighian tubule clones expressing Ras85DV12.Scer\UAS (using the MARCM system, under the control of Scer\GAL80αTub84B.PL and Scer\GAL4Scer\FRT.Act5C) is suppressed by co-expression of phlK497M.Scer\UAS.

Co-expression of phlK497M.Scer\UAS with Ras85DV12.Scer\UAS via Scer\GAL4esg-NP7397 results in a phenotype similar to that of Ras85DV12.Scer\UAS overexpression alone.

When phlK497M.Scer\UAS is driven by Scer\GAL4dpp.blk1 in a ftG-rv background a suppression is seen in the ftG-rv overgrowth phenotype in the eye disc, but no effect is seen in the wing disc. An increase in cell non-autonomous cell death is seen in these discs. When phlK497M.Scer\UAS is driven by Scer\GAL4ey.PH in ftG-rv mutant eyes, eyes are significantly smaller than wild-type. However eye bristle duplications are still seen.

Denticle belt fusions in the cuticles of rho7M43; ru1 double homozygous embryos are enhanced by phlK497M.Scer\UAS; Scer\GAL4prd.RG1.

phlK497M.Scer\UAS/wg-C.Scer\UAS double mutant clones (driven by Scer\GAL4en-e16E) covering the whole of the posterior compartment of the wing disc show a partial reduction in the size of the wing pouch and notum, and a suppression of the notum to wing transformations seen in phlK497M.Scer\UAS clones.
