Larval muscle 6/7 NMJs in AP-2σKG02457/AP-2σang-7, transheterozygotes show significantly more and significantly smaller boutons, which exhibit a significant decrease in periodic microtubule loop structures (Futsch staining), and show a significant decrease in the vesicle pool, as compared to controls. Electrophysiology recordingS at the NMJ also show significant increases in mEJP frequency and amplitude, and significant decreases in EJP amplitude and quantal content, and show a significant decline in EJP amplitude readings upon high-frequency stimulation at 34[o]C conditions even after a 4min rest period, as compared to controls.

AP-2σKG02457 mutant MARCM clones contain supernumerary neuroblasts, compared with control clones.

The expression of AP-2σUAS.cCa under the control of Scer\GAL4elav-C155 rescues the larval synaptic phenotypes but not the lethality observed in AP-2σKG02457 homozygotes and in AP-2σKG02457/AP-2σang-7 transheterozygotes. Expression under the control of Scer\GAL4Act5C.PU rescues both lethality and larval synaptic phenotypes in AP-2σKG02457 homozygotes and in AP-2σKG02457/AP-2σang-7 transheterozygotes.
