Nedd8AN015 mutant female germline stem cell clones are lost from the niche much more quickly than wild type controls.

Nedd8AN015 mutant Mushroom Body neuroblast clones consist of only small numbers of γ-type neurons, indicating severe defects in Mushroom Body neurogenesis. γ neurons homozygous for Nedd8AN015 fail to extend their axons to the midline in both neuroblast clones and single-cell/two-cell clones.

The majority of adult Nedd8AN015 mutant neurons possess three to five primary dendritic processes, comparable with wild-type. However, the averaged total dendritic length of mutant γ neurons is only 32.9υm, much shorter than the 61.3υm found in wild-type γ neurons. Approximately 42.5% of Nedd8AN015 mutant dendrites are extremely short and hardly separable from their primary neurites. In addition, terminal elaboration looks abnormal in mutant dendrites, often stalling with dense irregular endings.

Very few large homozygous clones are recovered in adults. Small clones are present in developing discs. Clones of cells in the anterior of the eye disc constrict in the apical surface precociously, indicating that they adopt morphogenetic furrow fate precociously.
