Description

A deletion of 5714 nucleotides from the 5' end of the inserted P{EP}, the exon 1 sequence 3' to the insertion site, intron 1, and the 5' 814 nucleotides of exon 2; the duplicated nucleotides 106--113 of exon 1 are followed by the 3' portion of the P-element, and nucleotide 2272 from its 3' end is contiguous with the codon for Pro250.

EndoAΔ4/EndoA26, EndoAS75D.CyPet larvae show increased autophagosomes (Atg8-mCherry) at NMJ boutons, under both fed and starved conditions.

In EndoA26/EndoAΔ4 transheterozygotes containing a copy of EndoAT:Avic\GFP-CyPet, the presynaptic region of third instar larval neuromuscular junctions does not present abnormal density of autophagosome (detected as fluorescent puncta of Atg8 or Lamp in confocal microscopy images) under normal diet, nor defects in starvation-induced formation of autophagosomes, as compared to controls. In EndoA26/EndoAΔ4 transheterozygotes containing a copy of EndoAS75A.T:Avic\GFP-CyPet, the presynaptic region of third instar larval neuromuscular junctions fails to present the expected starvation-induced formation of autophagosomes (detected as fluorescent puncta of Atg8 or Lamp in confocal microscopy images), but presents no defects in autophagosome density under normal diet, as compared to controls; the corresponding third instar larval effector neuron axons do not present defects in autophagosome density, as compared to controls. In EndoA26/EndoAΔ4 transheterozygotes containing a copy of EndoAS75D.T:Avic\GFP-CyPet, the presynaptic region of third instar larval neuromuscular junctions presents an abnormally high density of autophagosomes (detected as fluorescent puncta of Atg8 or Lamp in confocal microscopy images) under normal diet, but does not present defects in the starvation-induced autophagosome formation, as compared to controls.

EndoA26/EndoAΔ4 transheterozygous adults containing a copy of either EndoAS75A.T:Avic\GFP-CyPet or EndoAS75D.T:Avic\GFP-CyPet exhibit significant decreases in the number of dopaminergic PPM3 and PPL1 neurons, but not dopaminergic PPM1 and PPM2 neurons, at day 30, but not day 1, after eclosion, as compared to controls; in adults reared under constant light, but not under constant dark, the eye exhibits a significant decrease in the rhabdomere number per ommatidium and in the photoreceptor terminal number per lamina cartridge (at day 7 of light exposure), as compared to controls; the eye of individuals reared under either constant light or constant dark also exhibits a significant decrease in depolarization amplitude in response to light pulses (at day 3 of light exposure), as compared to controls.

Homozygous endoA26 causes lethality at the latest in second instar larvae. endoA10/endoA26 transheterozygotes show a strong loss of motility phenotype and lethality with paralysis during the L3 larval stage. No significant mEJP phenotype is seen in endoA10/endoA26 larvae, suggesting synaptic vesicle exocytosis is normal. However synaptic vesicle (SV) endocytosis is affected, about 40% of boutons exhibit a reduction in dye uptake of >30% (compared to about 3% of boutons in controls), and <5% of boutons exhibit high uptake (compared to about 15% in controls). endoA10/endoA26 larvae also show defects in the ultrastructure of neuromuscular junctions. IN about 30% of synapses the presynaptic terminal is almost completely depleted of synaptic vesicles (referred to as Grade III depletion). The few residual SVs are associated preferentially with the T-bar. i.e. the active zone. The plasmalamella of these synapses do not show an obvious accumulation of budding intermediates with a significant degree of membrane curvature. However, in the majority of cases (around 60%) there is substantial (but not an almost total) depletion of SVs (Grade II depletion), with the remaining being concentrated at the active zone. In ~10% of the synapses, there is no striking loss of SVs (Grade I depletion), but nonetheless an obvious change in ultrastructure, i.e. a remarkable increase in endocytic pits at the plasma membrane. Virtually all of these represent early-stage budding intermediates. In Grade I-III depleted synapses, there is an increased incidence of vacuoles, which are often found in close proximity to the plasma membrane and in the vicinity of te active zone. On average endoA10/endoA26 larvae show a an ~80% reduction in SVs and a three fold increase in the occurrence of early stage budding intermediates. About 55% of synapses are grade I and about 40% grade II depleted.
