UAS regulatory sequences drive expression of a chimeric construct consisting of the extracellular and transmembrane domains of kek1 fused to a portion of the cytoplasmic domain of Egfr (which includes the kinase domain but lacks the C-tail). The chimeric protein is tagged at the C-terminal end with GFP.
Expression of Egfr::kek1KEΔCG.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4CY2 results in eggs with dorsalised chorions. Expression of Egfr::kek1KEΔCG.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4GMR.PF results in a rough eye phenotype.
Egfr::kek1KEΔCG.UAS.GFP, Scer\GAL4CY2 has egg chorion phenotype, suppressible | partially by Df(2R)Egfr3/+
Egfr::kek1KEΔCG.UAS.GFP, Scer\GAL4CY2 has egg chorion phenotype, suppressible | partially by Egfrf8/Egfr[+]
Egfr::kek1KEΔCG.UAS.GFP, Scer\GAL4CY2 has egg chorion phenotype, suppressible by EgfrDN.UAS.cBa, Scer\GAL4CY2
Egfr::kek1KEΔCG.UAS.GFP, Scer\GAL4CY2 has egg chorion phenotype, suppressible by kek1UAS.cGa, Scer\GAL4CY2
Egfr::kek1KEΔCG.UAS.GFP, Scer\GAL4CY2 has egg chorion phenotype, non-suppressible by Egfr[+]/Egfrt1
Egfr::kek1KEΔCG.UAS.GFP, Scer\GAL4CY2 has egg chorion phenotype, non-suppressible by grk3/grk[+]
The dorsalised chorion phenotype caused by expression of Egfr::kek1KEΔCG.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4CY2 is completely suppressed by coexpression of kek1Scer\UAS.cGa or EgfrDN.Scer\UAS.cBa and is partially suppressed if the flies are also heterozygous for one of Df(2R)Egfr3 or Egfrf8. The dorsalised chorion phenotype caused by expression of Egfr::kek1KEΔCG.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4CY2 is not suppressed if the flies are also heterozygous for one of Egfrt1 or grk3.
