shepBG00836/shepBG00836 Burs-positive CCAP neurons have smaller cell bodies and fewer projections than heterozygous controls in pharate adults.

shepBG00836/Df(3L)ED210 mutant females display defects in courtship and mating behaviors. When crossed to wild type males the females vigorously kick at the males during mounting and copulation, and copulation duration is reduced compared to controls. Copulation latency (time taken to begin copulation) is unchanged from controls. In contrast to wild type, shepBG00836/Df(3L)ED210 females display extensive three-legged (compared to the normal two-legged) grooming and expulsion of seminal fluids from the reproductive tract after copulation.

At the P14 pharate adult stage, the bursicon neuron somata in shepBG00836/Df(3L)ED210 mutant animals have lost the multiangular morphology characteristic of that stage and become more rounded. Soma area and peripheral axon branching are reduced. The peripheral bursicon axons also display fewer boutons than controls. Bursicon neuron soma area is unchanged in shepBG00836/Df(3L)ED210 larvae compared to controls and there is no change in the number and size distribution of NMJ boutons.

At the P14 pharate adult stage, the bursicon neuron somata in homozygous shepBG00836 mutant animals are reduced in area. Bursicon neuron soma area is unchanged in shepBG00836/Df(3L)ED210 larvae compared to controls.

Expression of shepScer\UAS.EG under the control of Scer\GAL4BG00836 partially suppresses the kicking exhibited by shepBG00836/Df(3L)ED210 mutant females during mounting and copulation. The copulation duration is restored to wild type levels. Expression of shepScer\UAS.EG does not rescue the grooming defects seen after copulation.

Expression of shepScer\UAS.EG under the control of Scer\GAL4BG00836 fully rescues the bursicon neuron soma area, peripheral axon branch number and bouton number defects seen in shepBG00836/Df(3L)ED210 mutant pharate adults.
