Expression of sraScer\UAS.cEa under the control of Scer\GAL4GMR.PU results in a mild rough-eye phenotype compared to controls.

Overexpression of sraScer\UAS.cEa under the control of Scer\GAL4elav.PLu in a wild-type background does not affect activity or sleep.

Expression of sraScer\UAS.cEa in developing eyes, driven by Scer\GAL4GMR.PF, does not cause a phenotype. Expression of sraScer\UAS.cEa, under the control of Scer\GAL4en-e16E, does not cause a phenotype in the wing.

Expression of sraScer\UAS.cEa under the control of Scer\GAL4elav-C155 or Scer\GAL4sca-537.4 results in increased ovulation in virgin females compared to control virgin females. Within 30 minutes in a mating assay, only 50% of virgin females expressing sraScer\UAS.cEa under the control of Scer\GAL4elav-C155 have mated, compared to 82% of wild-type virgin females. Virgin females expressing sraScer\UAS.cEa under the control of Scer\GAL4elav-C155 elicit intermediate courtship index (CI) levels (CI is the period of time within the observation period that a male displays any element of courtship behaviour), that are between the level elicited by virgin wild-type females and mated wild-type females. Virgin females expressing sraScer\UAS.cEa under the control of Scer\GAL4elav-C155 have a high extrusion index (EI, the percentage of a time a female shows extrusion behaviour when being courted by a male) which is nearly comparable to that of wild-type mated females. Within 30 minutes in a mating assay, only 65% of virgin females expressing sraScer\UAS.cEa under the control of Scer\GAL4sca-537.4 have mated, compared to 82% of wild-type virgin females. Virgin females expressing sraScer\UAS.cEa under the control of Scer\GAL4sca-537.4 have a significantly higher extrusion index (EI, the percentage of a time a female shows extrusion behaviour when being courted by a male) than wild-type virgin females.

The severe small and rough eye phenotype observed in flies expressing Hsap\APPAβ42.Scer\UAS.cBa under the control of Scer\GAL4GMR.PU is exacerbated further by co-expression of sraScer\UAS.cEa.

Co-expression of sraScer\UAS.cEa driven by Scer\GAL4nos.UTR.T:Hsim\VP16 suppresses the embryonic lethality resulting from the overexpression of Pp2B-14DDN.Scer\UAS.P\T in the female germ-line.

The rough eye phenotype seen in flies that express Pp2B-14Dact.Scer\UAS under the control of Scer\GAL4GMR.PF is completely suppressed by coexpression of sraScer\UAS.cEa. Additionally, the wing phenotypes seen in flies expressing Pp2B-14Dact.Scer\UAS under the control of Scer\GAL4en-e16E are completely suppressed by sraScer\UAS.cEa coexpression.

Comments

Expression of sraScer\UAS.cEa in the nervous system under the control of Scer\GAL4elav.PLu restores the increased locomotor activity found in sraKO mutants back to wild-type levels. Similarly, sleep is completely restored to wild-type amounts in this genotype. Concurrent with this, a wild-type lifespan is also restored.
