Description

Excision of the P{EP} element, resulting in a 3.1kb deletion that removes the first exon of the S6kII gene (nucleotides 95440-98535 of AE003574) as well as some parts of the putative promoter region and the first intron.

NMJ size is significantly reduced in S6kIID1 mutants compared with wild type controls. The number and area of pre-synaptic active zones per NMJ and number and area of post-synaptic GluRIID fields are also reduced. The ratio between active zones and GluRIID fields is unchanged from wild type and no apposition defects are observed. Bouton number is increased, but they are smaller in size and contain fewer synaptic sites.

S6kIID1 flies are short lived and show a disarrayed eye structure with increased numbers of photoreceptor rhabdomeres, cone cells and pigment cells, which become more pronounced at higher temperatures, as compared to controls; these mutants show ectopic differentiation of retinal cells at both the pupal and the adult stage. S6kIID1 heterozygotes and hemizygotes, however, show normal eyes. Around 16% of S6kIID1 flies raised at 25[o]C show ectopic veins in various regions of the wing, which becomes more severe at 29[o]C, as compared to controls. S6kIID1/Df(1)R8A transheterozygotes display identical eye and wing phenotypes to S6kIID1 homozygotes.

The roughened eye and increased rhabdomere phenotypes seen when either phlScer\UAS.F179 or Ras85DV12.Scer\UAS is expressed under the control of Scer\GAL4hs.2sev are enhanced in a S6kIID1/+ or S6kIID1/Y background.
