Orct2^EP1072 embryos show the characteristic U-shape phenotype of embryos unable to retract the germ band.

Orct2^R106/Orct2^EP1072 adult flies do not show any growth defect when raised at 25^oC. When raised at 18^oC, Orct2^R106/Orct2^EP1072 flies are smaller than wild-type flies.

Selected as: a P{EP} insertion line that modifies the pnr^MD237/+ phenotype when expressed using Scer\GAL4^pnr-MD237.

Comments

The orientation of the P{EP}EP1072^EP1072 element is opposite to the direction of Orct2 transcription. Precise excision of the P{EP} element reverts the modifying effect of EP1072^EP1072 on the rough eye phenotype caused by expression of Hsap\MAPT^{V337M.Scer\UAS} under the control of Scer\GAL4^GMR.PF.

Although this insertion is somewhat upstream of the Orct2 gene, Orct2 mRNA expression is reduced in flies carrying the insertion and Orct2 expression can be driven using the Scer\GAL4^Orct2-R107 driver. The phenotype of Orct2^EP1072 mutants is similar to mutants carrying other Orct2 alleles.
