Knockdown of Itp-r83AGD1676 (along with UAS-Dcr-2 expression) in the prothoracic gland (driven by Scer\GAL4phm.PO) leads to growth arrest during larval development (approximately 30% in the first or second stage and 10% in the third instar stage). Arrested third instar larvae also show an overgrowth phenotype due to the extended larval feeding period; feeding ecdysone (E) to these larvae almost fully rescues the arrest phenotype. Those larvae that do initiate pupariation are significantly delayed in doing so and exhibit overgrowth phenotypes (rescued by E feeding). Scer\GAL4phm.PO>Itp-r83AGD1676 larvae (during the extended third instar larval stage) show an accumulation of Syt1Scer\UAS.T:Avic\GFP-EGFP vesicles in the cytoplasm (especially adjacent to the plasma membrane). Scer\GAL4phm.PO>Itp-r83AGD1676 larvae show normal gross morphology of the prothoracic gland.

Expressing Itp-r83AGD1676 in the fat body under the control of Scer\GAL4ppl.PP increases lipid droplet size.

Expression of Itp-r83AGD1676 results in a significant decrease in the number of dendrite branch terminals in either class I ddaD and ddaE (when expression is driven by Scer\GAL4221, in combination with Dicer-2 for efficient RNAi) or class IV ddaC (when expression is driven by Scer\GAL4ppk.PG, in combination with Dicer-2 for efficient RNAi) abdominal dorsal multidendritic neurons from third instar larvae, as compared to controls.

Adults expressing Itp-r83AGD1676 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4pnr-MD237 results in a colour difference between the central Scer\GAL4pnr-MD237 expression domain of the notum and the surrounding lateral region in 0% or 80-90% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.
