Expression of shgGD14421 under the control of Scer\GAL4e22c results in abnormal large syncytia formation in the epidermis of third instar larvae, although the penetrance is low (less than 20% larvae).

Knockdown of shg in the outer, migratory border cells, through expression of shgGD14421, under the control of Scer\GAL4slbo.2.6, in contrast to wild-type, results in a significant reduction of direction persistence in migration, with clusters ending up in ectopic locations, and the migration paths wandering, rather than migrating in a straight line. Though these cells are still motile and remain clustered, shg knockdown reduces the average speed from 0.67 υm/min to 0.26 υm/min.

Animals expressing shgGD14421 under the control of Scer\GAL4Mef2.PR have missing flight muscles.

Adults expressing shgGD14421 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in flightless flies or early pupal lethality.

Expression under the control of Scer\GAL4Mef2.PR results in missing adult indirect flight muscles.

The phenotype of missing flight muscles that is seen in animals expressing shgGD14421 under the control of Scer\GAL4Mef2.PR is not rescued if they are carrying the P{FlyFos-045685} construct (which contains D. pseudoobscura pseudoobscura fosmid sequence that includes the region orthologous to shg), indicating either that the P{GD14421} RNAi phenotype is unspecific, or that the D. pseudoobscura shg ortholog is also targeted by the P{GD14421} hairpin.
